    My story is not very different from biologists who graduated with Ph.D’s in the late 90’s or early 00’s. Big data was something restricted to the Physics department, in the form of a mainframe machine that usually took up a whole room behind a locked door and relatively few onsite terminals. Very few biologists like myself walked through that door.

In this session, we will address some of the key issues around reproducibility and the practical steps that can be taken to reduce creeping sources of error.

    With the advent of single cell analyses many unanticipated cell biological phenomena have been discovered including, new classes of RNA, identification of RNA targeting sequences and perhaps most surprisingly the discovery of cell-to-cell variation in transcriptome abundances even in presumptively identical cells.

    I will discuss our published experimental approach to formally test whether aberrant DNA methylation can actually cause cancer in mice in vivo. In addition, I will talk about our ongoing studies, and share our latest results to test novel strategies for epigenetic preventive and therapeutic interventions.

    The ability to conduct saturating genetic knockout screens in human cells with CRISPR-Cas9 is catalyzing a revolution in functional genomics and cancer discovery.     By attending this session, it is my goal that each participant will learn the following pieces of information: 

    - There will be a basic review of CRISPR-Cas adaptive immunity and nomenclature.

    - Class 2 Cas proteins are applied for genome engineering or other applications because they require a single protein and are simpler than utilizing a Class 1 CRISPR-Cas protein complex.
